Source text - Persian (Farsi) داروهای ژنتیکی برای درمان بیماری¬های ناشی از نقص¬های کروموزومی چندین دهه است که توسط محققین مورد استفاده قرار می¬گیرد. موضوع اصلی تحقیقات در این حوزه که به ژن درمانی شناخته شده است جستجوی دستیابی به روش¬های تحویل ایمن، موثر و قابل کنترل است. وکتورهای ویروسی بویژه وکتورهای ادنوویروس، رتروویروس، لنتی ویروس که از سال 1990 برای ژن درمانی مورد استفاده قرار گرفتند کارایی¬های انتقال بالایی را نشان می¬دادند اما مسائل مرتبط با ایمنی استفاده از این وکتورها را با مشکل روبرو ساخت و توجه محققین را به استفاده از سیستم¬های تحویل جایگزین سوق داد. با ورود نانوتکنولوژی و امکان بهبود ویژگی¬های نانومواد دسته¬ی دیگری از حامل¬های تحویل که به وکتورهای غیرویروسی(سنتزی) شناخته¬می¬شوند مورد توجه قرار گرفتند. این وکتورها که با جاذبه¬های الکتروساتیک به DNA یا RNA وصل می¬شوند مواد ژنتیکی را فشرده، حفاظت و به درون سلول وارد می¬نمایند. وکتور مورد نظر برای انجام چنین کارهایی باید دارای ویژگی¬هایی مثل بار مثبت سطحی کافی، زیست¬سازگاری و ظرفیت بافری بالا برای ورود به سلول و رهاسازی محموله¬مورد نظر باشد. در میان وکتورهای غیرویروسی لیپیدها و پلیمرها مکررا به عنوان معرف ترنسفکشن مورد استفاده قرار می¬گیرند. پلیمرهای کاتیونی که دارای آمین¬های نوع اول، دوم، سوم، چهارم و گروههای با بارمثبت دیگری مثل آمیدها هستند ویژگی¬های مود نیاز یک وکتورر را دارند. از جمله این پلیمرها می¬توان پلی اتیلن ایمین، پلی لیزین و پلی¬آمیدوآمین¬ها را نام برد. در این میان مسئله¬ی بعدی که در انتخاب یک وکتور باید به آن پرداخته شود زیست سازگاری است در میان وکتورهایی که تاکنون به عنوان معرف ترنسفکشن مورد استفاده قرار گرفتند دندریمرهای پلی¬آمیدوآمین کمترین میزان سمیت و بالاترین زست سازگاری را نشان می¬دهند. دندریمرها می¬توانند کمپلکس¬های قابل حل و پایداری با DNA برقرار کنند و همچنین مزیت دیگز آنها این است که رنج غلظتی گسترده¬ای بین ترنسفکشن و سمیت آنها وجود دارد.
Translation - English Researchers have been using genetic drugs for the treatment of diseases caused by chromosomal defects for decades. The main subject of research in this field, known as gene therapy, has been the search for safe, effective and controllable delivery methods. Viral vectors, in particular, adenoviral, retroviral, and lentiviral vectors that have been in use for gene therapy since 1990 have shown a high transmission efficiency, but safety issues associated with the use of these vectors led researchers to consider alternative delivery systems. With the advent of nanotechnology and the possibility to improve the characteristics of nanomaterials, other sets of delivery carriers known as non-viral (synthesized) vectors were introduced.
These vectors that attach to DNA or RNA by electrostatic force, could compress, protect and deliver genetic material into cells. To be able to perform such tasks, vectors should have proper features such as a positively charged surface, biocompatibility, and high buffering capacity to enter the cell and release the desired cargo. Among non-viral vectors, lipids and polymers are frequently used for transfection. Cationic polymers that contain first, second, third, or fourth amins or/and positively charged groups such as amides, have the required characteristics to play the role of a vector. Among these polymers, polyethyleneimine, poly-L-lysine, and poly-amidoamine can be named. The next issue that must be addressed in selecting a vector is its biocompatibility. Among vectors that have been used as a transfection agent hitherto, Polyamidoamine (PAMAM) dendrimers have shown the least amount of toxicity and the highest biocompatibility. Dendrimers can make soluble and stable complexes with DNA. The other advantage of dendrimers is that there is a wide range of concentrations between their toxicity and transfection ability.
